Lecture:
The lecture covers basic molecular techniques including vectors and cloning strategies, promoter analyses, DNA-Protein interactions, Biocomputing for the analysis of sequences, genes and genomes, hybrid dysgenesis, generation of transgenic organisms, principles of transgenes and their application, legislature for the safe handling of genetically modified organisms (MGO).
Seminar:
Readings of current methods in molecular genetics; solutions of problems in molecular genetics in small groups with the help of bioinformatic tools
Practical course:
- Safety procedures and good laboratory standards in the molecular genetics lab
- Gene mapping, Southernblot, stringency, detection
- Transformation of bacteria, competence, efficiency, selection
- Expression vectors and constructs, bacterial protein expression, purification of fusion proteins by chromatography, PAGE
- Genotyping of transgenic lines, PCR, primer selection, diagnostic
- Protein-protein interactions in the yeast two-hybrid system
- Immunohistochemical detection of ectopic protein induction
- Gene manipulation: tissue specific overexpression of transgenes
- Temporally and spatially restricted RNA interference for knock down of gene acticity
- Quality management in the design and execution of molecular biology experiments (controls and documentation) - Comments:
